The closure, part of a series of planned disruptions on the A282 this week, will take place from 10pm to 5am, with southbound traffic diverted through the East Tunnel under a contraflow system.
The closure is expected to cause significant delays for drivers, particularly those traveling late at night or early in the morning.
Other closures at the Dartford Crossing, including the West Tunnel and slip roads, are also scheduled throughout the week, with diversions via National Highways routes in place.
